Pharmacological Action

Etoricoxib: Selective COX-2 inhibitor — reduces prostaglandin synthesis, providing potent analgesic and anti-inflammatory effects with reduced GI risk. Thiocolchicoside: Central and peripheral muscle relaxant — acts on GABA-A and glycine receptors to relieve skeletal muscle spasm.

Mechanism

Etoricoxib selectively blocks the COX-2 enzyme, inhibiting the conversion of arachidonic acid to pro-inflammatory prostaglandins, thereby reducing pain and inflammation at the source without significantly affecting COX-1 (gastro-protective). Thiocolchicoside targets inflammation-driven spasm by modulating inhibitory neurotransmitter receptors (GABA-A & glycine), leading to muscle relaxation, restored mobility, and reduced chronic stiffness.
